摘要
目的观察辛伐他汀对高脂血症大鼠主动脉平滑肌细胞凋亡的影响。方法采用高脂饲料喂养的方法建立高脂血症大鼠模型,将普通饲料喂养的SD大鼠设为正常对照组7只,成模大鼠随机分为高脂血症组10只及辛伐他汀组10只,药物干预4周后,处死大鼠取主动脉测定平滑肌细胞凋亡率。结果与高脂血症模型组比较,辛伐他汀组甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)均明显下降(P<0.05或P<0.01)。与高脂血症模型组比较,辛伐他汀组肝细胞脂肪变性的程度明显减轻。辛伐他汀组主动脉平滑肌细胞凋亡率为(34.8±1.86)%,高于高脂血症组主动脉平滑肌细胞凋亡率(27.1±1.37)%(P<0.01)。结论辛伐他汀能够降低高脂血症大鼠血脂,增加高脂血症大鼠平滑肌细胞凋亡率,延缓动脉粥样硬化的发生。
Objective To explore the effect of simvastatin on smooth muscle cells apoptosis of aorta in hyperlipidemic rats. Methods The model of hyperlipidemic rats was induced by high - fat diet feeding and 7 SD rats with normal diet feeding were the normal contral group, hyperlipidemic model rats were randomly divided into the hyperlipidemic groop and simvastatin groups. The course of treatment lasted for 4 weeks and all rats were sacrificed later. Aortas were taken for measuring the index of smooth muscle ceils apoptosis. Results Compared with control group, the results of triglyceride ( TG), cholesterol ( TC ) and low density lipoprotein (EDL) in simvastatin group were lower than those in the hyperlipidemic groop(P 〈0.05 or P 〈0.01 ). Hepatic steatosis of simvastatin group were significantly reduced compared to that in the hyperlipidemic groop. The level of the index of smooth muscle cells apoptosis in the simvastatin group was higher than that in the hyperlipidemic groop(34.8 ± 1.86% vs 27.1 ± 1.37%, P 〈 0. 01 ). Conclusion Simvastatin can decrease the levels of the blood lipid and increase the levels of the index of smooth muscle ceils apoptosis of hyperlipidemic rats, slow down the atherosclerosis of hyperlipidemic rats.
