摘要
研究羊肚菌Ydj0705的液体发酵培养基配方,确定液体发酵的工艺条件及方法,开发真菌资源菌丝产品。采用正交设计法,优化羊肚菌Ydj0705培养基配方和培养条件,从而建立羊肚菌Ydj0705液体发酵工艺。结果显示,羊肚菌Ydj0705液体发酵培养基最佳配方为:玉米粉30g/L,葡萄糖10g/L,黄豆粉20g/L,酵母粉5g/L,KH2PO4和MgSO4适量;最优液体发酵培养条件为:培养温度26℃,摇床转速定量,培养基初始pH为6.0,装液量为100mL培养液,接种量10%,发酵时间144h。羊肚菌Ydj0705液体发酵工艺能高效生产菌丝体,为开发羊肚菌真菌资源产品奠定了基础。
This study was to study the liquid fermentation medium compounding of Morchella esculenta(Ydj0705), to optimize the liquid fermentation condition, and process with liquid fermen- tation by the orthogonal design. Five strains were screened and the optimum liquid fermentations conditions of Morchella esculenta(Ydj0705) were established. The optimal culture medium was: corn power 30 g/L, glucose 10 g/L, soybean meal 20 g/L, yeast extract 5 g/L,right amount of KH2PO4 and MgSO4. Results of the test show that the optimal culture conditions was as follows: temperature 26~C, quantitative rotation speed, pH6 . 0, the liquid volume 100 mL/250 mL, inoculation rate 10%, 6 days for the end time of fermentation. The best liquid fermentation of Morchella esculenta(Ydj0705) increased the cell concentration greatly, accelerated the mushroom output, and laid the foundation for the subsequent development of products of cultured or natural fungi resources.
