摘要
目的 :探讨Gd DTPA脂质体造影剂对大鼠肝癌灶的诊断价值。方法 :采用冻融超声法制备出粒径 0 .61±0 .3 4 μm的装载Gd DTPA的脂质体 ,二乙基亚硝胺法建立大鼠肝癌灶模型 ( 5例 ) ;观察静注 0 .1mmol/10 0 g体重Gd DT PA脂质体后大鼠肝癌灶显示情况 ,并与尸检对照。结果 :脂质体造影剂包裹率为 2 0 .1% ,能有效、持续地造成肝脏选择性靶向强化 ,从而衬托出肝癌病灶 ,使癌灶检出率由平扫 17.6%上升至增强后的 76.5 % ( 0 .0 1<P <0 .0 5 )。结论 :顺磁性脂质体能有效地提高肝脏造影效果 。
Objective:To investigate the value of liposomal Gd-DTPA enhanced MR imaging for improving detection of small hepatocellular carcinoma nodules (HCCs) in rat model.Methods:Gd-DTPA liposomes,which averaged 0.61±0.34μM in diameter,were synthesized by freeze-thaw-sonification extrusion process.The rat model with HCCs was chemically induced by oral administration of diethylnitrosamine (DENA) for 16 weeks (n=5).After intravenous injection of Gd-DTPA-liposomes at a dose of 0.1mmol per 100g body weight,MR contrast features of HCCs were observed and correlated with pathologic changes.Results:Encapsulation efficiency was 20.1% in the present liposomal Gd-DTPA preparation.After the injection,liposomal Gd-DTPA resulted in a persisting and considerable liver parenchymal contrast enhancement,and permited significant improvement of detection of HCCs.The detecting rate of HCCs was 17.6% on precontrast T 1 weighted images,while rised to 76.5% on the postcontrast images (0.01< P <0.05).Conclusions:Paramagnetic liposomes can markedly increase hepatic uptake and result in persisting liver enhancement.It has high values for improving detection of tiny HCC nodules.
