摘要
目的探讨多次低剂量x线照射对小鼠精子DNA完整性的影响。方法采用0、O.025、0.075及0.100GyX线多次照射小鼠,单细胞凝胶电泳技术(SCGE)对小鼠精子DNA完整性进行分析。结果随着单次照射剂量的增大,小鼠精子DNA的损伤级别增高;0.025、0.075及0.100Gy照射组小鼠精子DNA Olive尾距、精子DNA彗星尾长及精子DNA尾部DNA含量百分率分别与对照组比较差异具有显著统计学意义(P〈0.05),且随着单次照射剂量的增大差异越明显。结论多次低剂量x线全身照射小鼠可影响精子DNA完整性,且随着单次照射剂量增加,损伤加重。
Objective To investigate the effects of multiple low-dose X-ray irradiation on mouse sperm DNA integrity. Methods The mice were irradiated repeatedly by low-dose of 0, 0.025, 0.075 and 0.100Gy X-ray respectively, and its sperm DNA integrity was detected by Single Cell Geleletrophoresis (SCGE). Results The injury grade of mouse sperm DNA was escalated along with increase of the single irradiation dose. There were significant differences in sperm DNA Olive tail moment, comet tail length and percentage of tail DNA content between the three groups (0.025 Gy, 0.075Gy, 0.100Gy) and the control group (P〈0.05). The difference was markedly seen along with increase of the single irradiation dose. Conclusion Sperm DNA integrity was damaged by repeated irradiation with low-dose X-ray, and the irradiation injury level elevated along with the increasing dose in single irradiation.
