摘要
目的 探讨卵巢癌细胞在经多肿瘤抑制基因(MTS1)转染后其超微结构及单个细胞增殖力的变化 .方法 采用软琼脂克隆形成法 ,对比性观察卵巢癌细胞 HO- 8910在转染 MTS1后单个细胞增殖力的改变 ,并应用透射电镜观察其细胞超微结构的变化 .结果 卵巢癌细胞 HO- 8910在转染 MTS1后其单个细胞的增殖力明显下降 ,而转染空载体未产生任何影响 ;同时电镜结果示细胞在转染 MTS1后其超微结构亦出现趋向死亡的征象 .结论
AIM To study the transformation of ultrastructures and proliferation of human ovarian cancer cell line (HO 8910) transfected by MTS1. METHODS By the assay of soft agar medium, the cloning efficiency and proliferation of a single cell were compared among the HO 8910, 8910 pcDNA3 and 8910 p16 cells. The ultrastructures of them were also observed under transmission electron microscope. RESULTS The cloning efficiency and proliferation of a single cell descended obviously after being transfected by MTS1. There was no difference between the HO 8910 and 8910 pcDNA3 cells. The results observed under transmission electron microscope showed that following transfection the ultrastructures in 8910 p16 tended to show signs of death. CONCLUSION By affecting the ultrastructures of HO 8910, MTS1 can downregulate the cloning efficiency and inhibit the extra growth of ovarian cancer cells.
