目的 探讨多癌基因反义寡核苷酸对慢性髓细胞白血病 (CML )的作用及其相互作用的影响 .方法 用台盼蓝拒染、集落形成、逆转录 -多聚酶链反应及流式细胞仪技术研究反义寡核苷酸对 K5 6 2细胞株和 CML 骨髓细胞的作用 .结果 c- myb反义寡核苷酸通过促进 CML 细胞凋亡 ,发挥对 bcr/abl反义寡核苷酸的协同作用 .结论 多癌基因反义核酸的联合应用可为
AIM To investigate the synergistic effects of bcr 3/ abl 2 and c myb antisense oligodeoxynucleotides (ASO) on chronic myeloid leukemia (CML). METHODS The synergistic effects of bcr 3/ abl 2 and c myb antisense oligodeoxynucleotides on 15 CML patients and the leukemic cell line K562 were studied by using trypan blue exclusion, formation of granulocyte macrophage colony (CFU GM), reverse transcription polymerase chain reaction (RT PCR) and flow cytometer (FCM) techniques. RESULTS The effects of ASO b/a on CML cells were observed by inducing apoptosis in CML cells. The synergistic effect of ASO m was exerted by enhancing apoptosis in CML cells. CONCLUSION The application of antisense oligodeoxynucleotides of mutiple oncogenes may provide a new way to cure CML.
