摘要
目的:探讨丝裂原活化蛋白激酶(MAPK)在慢性粒细胞白血病(CML)发病中的作用及其作用机理。方法:用脂质体转染法将MAPK反义寡核苷酸(ASO)导入CML细胞中,通过细胞集落形成、蛋白质含量、MAPK活性和MAPK表达的变化检测MAPK ASO对CML细胞的作用。结果:MAPK ASO可明显抑制细胞集落形成、蛋白质含量、MAPK活性和MAPK表达,其抑制率(%)分别为41.6、46.4、50.0和43.2,与对照组比较有显著差异(P<0.05)。结论:MAPK在CML发病中起重要,有可能成为治疗CML的新靶点。
Objective. To study the role of mitogen - activated protein kinase(MAPK) in pathogenesis of chronic myeloid leukemia (CML). Methods. After MAPK antisense oligodeoxynucleotide(ASO) was introduced into CML cells by liposomal transfection, the effects were evaluated by cell colony formation, MAPK protein content, MAPK activity and MAPK expression. Results: The cell colony formation, MAPK protein content, MAPK activity and MAPK expression were significantly inhibited by MAPK ASO, the inhibitory rates were 41. 6%, 46. 4%, 50. 0% and 43. 2%, respectively (P 〈 0.05). Conclusion: MAPK plays an important role in the pathogenesis of chronic myeloid leukemia and MAPK may become a new target in the treatment of CML.
