摘要
目的:研究褪黑素(MT)对细菌脂多糖(LPS)致宫内感染胎鼠脑组织TNF-α的表达变化,探讨MT对宫内感染胎鼠脑损伤的保护作用。方法:选用妊娠第19天SD大鼠随机分组:对照(S)组、宫内感染(LPS)组和MT处理组,腹腔注射LPS建立大鼠宫内感染模型,LPS组和S组分别腹腔注射LPS 500μg/kg和等容积生理盐水,MT组同时腹腔注射LPS500μg/kg+MT 10 mg/kg。分别于注药后1、6、12、24、48和72 h剖腹取胎鼠,RT-PCR技术检测胎鼠脑组织中TNF-αmR-NA的表达水平。结果:LPS组各小组较S组胎鼠脑组织中TNF-αmRNA含量均明显上升,LPS组在6h达峰值之后缓慢下降;MT组各小组TNF-αmRNA含量均低于LPS组,随时间延长MT组各小组逐渐上升,48 h达峰值,后稍下降。结论:TNF-α表达增加是LPS致宫内感染胎鼠脑损伤的机制之一;MT抑制TNF-α表达,对胎鼠脑损伤有保护作用。
Objective:To research the effect of melatonin on change of tumor necrosis factor-α(TNF-α) expression in fetal rat brain tissue suffered from intrauterine infection induced by lipopolysaccharide(LPS),and explore the protective effect of melatonin for brain injury of fetal rats with intrauterine infection.Methods:SD rats at 19 days of pregnancy were randomly divided into control group,LPS group,and melatonin group,the rats in LPS were treated with intraperitoneal injection of LPS(500 μg/kg),the rats in control group were treated with intraperitoneal injection of normal saline(500 μg/kg),and the rats in melatonin group were treated with intraperitoneal injection of LPS(500 μg/kg) and normal saline(10 mg/kg).Fetal rats were obtained by laparotomy at 1,6,12,24,48,and 72 hours after injection,RT-PCR was used to detect the expression level of TNF-α mRNA in fetal rat brain tissue.Results:The contents of TNF-α mRNA in subgroups of LPS group were statistically significantly higher than those of control group,which decreased gradually after peak at 6 hours after injection;the contents of TNF-α mRNA in subgroups of melatonin group were statistically significantly lower than those of LPS group,which increased gradually with time extension,peaked at 48 hours,then decreased slightly.Conclusion:The increase of TNF-α expression is one of the mechanisms of brain injury of fetal rats with intrauterine infection induced by LPS;melatonin inhibits TNF-α expression can plays a protective role in brain injury of fetal rats.
