摘要
以蚱蝉(Cryptotympana pustulata)若虫为材料,采用正交试验法,对蚱蝉若虫蛋白质的提取条件进行了研究。结果表明,碱提蛋白法的蛋白质得率最高,其次为盐提蛋白法,Tris-HCl提蛋白法得率最低。碱提蛋白法的主要工艺指标为:碱液质量分数0.5%、温度60℃、时间60min、固液体积比为1∶5时蛋白质得率有最大值;盐提蛋白法的主要工艺指标为:盐液质量分数1.5%、温度40℃、时间120min、固液体积比为1∶10时蛋白质得率有最大值;Tris-HCl提蛋白法的主要工艺指标为:Tris-HCl液pH值8.1、抽提温度20℃、抽提时间30min、固液体积比1∶10时蛋白质得率有最大值。
The extraction methods of the protein from the larva of Cryptotympana pustulata was investiga- ted by the orthogonal test. The results showed that alkaline extraction method had the highest yield, fol- lowed by salt extraction and Tris-HCl extraction. The optimum extraction conditions for alkaline method with the highest protein extraction yield were obtained as alkali concentration 0.5%, temperature 60℃, extraction time 60 min, and the ratio of solid to liquid 1 : 5. The optimal conditions for salt method were obtained as salt concentration 1.5%, temperature 40℃, extraction time 120 min, and the ratio of solid to liquid 1 ~ 10. The best conditions of Tris-HC1 method were obtained as pH 8.1, temperature 20℃, extrac- tion time 30 min, and the ratio of solid to liquid 1 : 10.
