摘要
目的体外研究鼠尾皮肤成纤维细胞(MTSF)发生老化时,ANGPTL-1中的表达及意义。方法分别收集不同年龄原代鼠尾皮肤成纤维细胞,并检测老化细胞的COL1、COL3及透明质酸合成量。用RT-PCR检测两组细胞中ANGPTL-1基因及胶原蛋白与透明质酸含量。结果 ANGPTL-1基因在青年组鼠尾皮肤成纤维细胞中的表达明显高于老年组(P<0.01);老年组成纤维细胞COL1、COL3及透明质酸合成量显著低于青年组成纤维细胞(P<0.01)。结论ANGPTL-1基因可能是延缓鼠尾皮肤成纤维细胞老化的重要基因之一,可应用于皮肤抗衰老的研究。
Objective To explore the expression of ANGPTL-1 gene, collagen and hyaluronic acid (HA) in skin fibroblast of aging mouse tail in vitro. Methods Primary mouse skin fibroblasts were collected in young and old mouse group. RT-PCR was used to measure expression of collagen and hyaluronic acid. Results RT-PCR assay showed that the express of ANGPTL1 is higher in young group than in older group. There was significant difference between 2 groups (P〈0.01). Conclusion ANGPTL-1 may be used as an effective factor to delay skin aging.
