摘要
目的:探索和建立人衰老皮肤成纤维细胞的体外培养的方法,为其在医学美容抗衰老研究中的应用提供科学依据。方法:植块法原代培养人衰老皮肤成纤维细胞;倒置显微镜下观察细胞的生长状态;HE染色观察细胞的形态及着色情况。结果:用植块法培养的人衰老皮肤成纤维细胞在显微镜下呈长梭形,有突起,平行、放射状或漩涡状排列。结论:确立了人衰老皮肤成纤维细胞的体外培养的方法,使其可供于延缓皮肤衰老的实验研究。
Objective To investigate a method for primary culture of aged human skin fibroblasts in vitro and to provide a reliable basis for researches of delaying skin aging. Methods Fibroblasts were cultured with explanted culture method. The morphology of fibroblasts were observed under inverted phase-contrast microscope and light microscope stained by HE. Results Aged human skin fibroblasts were obtained. Cells are tong spindle in shape arranged in parallel or radiate or swirl way under microscope. Conclusions The method for primary culture aged human skin fibroblast in vitro is found. This study provides an efficient technique for delaying skin aging research.
