摘要
【目的】研究中药益气血方对超促排卵小鼠卵巢生长分化因子-9(growth differentiation factor-9,GDF-9)与GDF-9B表达的影响。【方法】选用昆明种小鼠接受促性腺激素释放激素类似物(GnRHa)与孕马血清(PMSG)超促排卵,中药组予以益气血方(经后增殖颗粒)(剂量为1.5 mg/kg)灌胃,对照组予以生理盐水灌胃。注射绒毛膜促性腺激素(hCG)后取卵巢组织,采用免疫组织化学方法检测GDF-9与GDF-9B的表达与半定量分析,采用实时荧光定量PCR方法检测GDF-9、GDF-9B mRNA的含量。【结果】卵母细胞与颗粒细胞中检测到GDF-9的表达,GDF-9B未表达于卵母细胞,而在颗粒细胞中高表达。半定量分析结果显示中药组卵巢组织中GDF-9与GDF-9B蛋白表达均显著高于对照组(P<0.05),实时荧光定量PCR结果显示中药组GDF-9、GDF-9B mRNA表达水平均显著高于对照组(P<0.05)。【结论】中药益气血方可促进超促排卵小鼠卵巢GDF-9与GDF-9B的表达,可能通过此机制促进卵母细胞发育潜能,从而促进胚胎种植。
Objective To study the effect of Yiqixue Recipe, a Chinese prescription with the action of tonifying Qi and blood, on the expression of growth differentiation factor 9 (GDF-9) and growth differentiation factor 9B ( GDF- 9B) in controlled ovarian hyperstimulation (COH) mice. Methods Kunming mice were given gonadotrophin- releasing hormone agonist (GnRH) and pregnant mare serum gonadotrophin (PMSG) for controlled ovarian hyperstimulation. Mice in the Chinese medicine group were administrated intragastrically with Yiqixue Recipe (1.5mg/kg), and mice in the control group were given normal saline intragastrically. Ovarian tissues were collected at the ovulation stage after injection of human chorionic gonadotropin (hCG), and the expression and semi-quantitation of GDF-9 and GDF-9B were detected by immunohistochemical method. GDF-9 and GDF-9B mRNA content was measured by real-time fluorescence quantitative PCR. Results GDF-9 was detectable in oocytes and granular ceils by immunohistochemistry, and GDF-9B was detectable in granular cells but not in oocytes. The protein and mRNA levels of GDF-9 and GDF-9B were higher in Chinese herbal group than those in the control group (P 〈 0. 05) . Conclusion Yiqixue Recipe promotes the expression of ovarian GDF-9 and GDF- 9B in COH mice, and its possible mechanism is related to promoting the development potentiality of oocytes, and thus promoting embryo implantation.
