摘要
以藏药余甘子体外药效为指标,采取正交设计试验优选得到藏药余甘子药材抗肿瘤活性成分的最佳提取工艺,即以10倍量70%乙醇回流提取(2 h×2)。并建立了HPLC多波长融合法定量测定余甘子抗肿瘤活性成分中的没食子酸(1)、表儿茶素(2)和鞣花酸(3)。采用C18色谱柱,以0.2%磷酸-乙腈为流动相,梯度洗脱,检测波长分别为230 nm(1)、254 nm(2)和280 nm(3)。三者的线性范围分别为0.88~5.28 g、0.30~1.80 g和0.28~1.68 g。平均回收率分别为98.5%、98.6%和98.2%,RSD分别为1.02%、1.00%和1.06%。
The extraction of the antitumor constituents from Tibetan medicinal herb Phyllanthus emblica L. was optimized by orthogonal design with the in vitro efficacy as the index. The optimal process was as follows: 10 times of 70 % ethanol for 2 times, 2 h for each time. An HPLC method with muti-wavelength fusion was established for the determination of gallic acid (1), epicatechin (2) and ellagic acid (3) in antitumor constituents of Phyllanthus emblica L.. A C18 column was used with the mobile phase of 0.2 % phosphoric acid-acetonitrile by gradient elution at the detection wavelengths of 230 nm (1), 254 nm (2) and 280 nm (3). The calibration curves were linear in the ranges of 0.88 - 5.28 μg, 0.30 - 1.80μg and 0.28 - 1.68μg. Their average recoveries were 98.5%, 98.6% and 98.2%, with RSDs of 1.02 %, 1.00 % and 1.06 %, respectively.
