摘要
【 目的建立反相高效液相色谱测定血浆中茶碱浓度的方法。方法以ODS—C18柱为固定相,甲醇:水(32:68)为流动相;流速0.8ml/min;柱温35℃;检测波长275nm,测定茶碱的血药浓度。血浆中的茶碱用氯仿-异丙醇(95:5)提取。结果茶碱在4.0—24.00μg/ml浓度范围内呈良好线性关系,r=0.9994;平均回收率为99.06%;日内和日间RSD均小于2.0%。结论本法灵敏简便、准确可靠,可作为茶碱血药浓度监测的常规方法。
Objective To establish a method of RP - HPLC for the determination of theophylline concentration in human plasma. Methods RP - HPLC was carried out on Yilite - C18. The mobile phase was composed of metha- nol -water(32:68 ), the flow rate was l. 0 ml/min, the column temperature was set at 35℃, the detecting wavelength was set at 275nm. The wavelength for detection was 275 nm. The theophylline was extracted from plasma using a mix- ture of trichloromethane : isopropanol ( 95 : 5, V/V). Results The assay was shown to produce linear calibration curves over the range from 4.0 - 24.00μg/ml for theophylline, r = 0. 9997. The average recovery was 99.06% and the inter - day RSD and intra - day RSD of theophylline were less than 2.0%. Conclusion The method is found to be sensi- tive, simple, convenient'and accurate for the determination of theophylline concentration in human plasma. It also can be used as a routine method by TDM.
