摘要
目的:研究黄芪多糖对胃癌细胞MKN45的生长抑制作用及细胞周期的影响。方法:用RPMI 1640培养液对人胃癌细胞MKN45进行传代培养,采用MTT法检测黄芪多糖对MKN45的生长抑制作用,采用流式细胞术检测黄芪多糖对MKN45细胞周期的影响。结果:MTT法实验显示黄芪多糖可抑制MKN45细胞增殖;细胞周期分析提示黄芪多糖可将细胞MKN45阻滞于G0/G1期,效应均呈浓度和时间依赖性。结论:黄芪多糖能抑制胃癌细胞增殖,其机制之一是影响细胞周期使之阻滞于G0/G1期。
Objective :To observe the effects of Astragalus Polysaccharide on proliferation and ceil cycle of Human Gas- tric Carcinoma Cell Line MKN45. Methods:Subculture was carried on by Human Gastric Carcinoma Cell Line MKN45 in RPMI 1640 culture solution. MTT assay was used to determine the inhibitory effect of Astragalus Polysaccharide on the growth of MGG - 803 ceils. The changes of cell cycle of MKN45 were detected by flow cytometry. Result : MTT assay showed that Astragalus Polysaccharide could inhibit the proliferation of cell line MKN45, cell cycle analysis indicated that Astragalus Polysaccharide can arrest MKN45 at G0/GI phase, both of them were time - and dose - dependent manner. Conclusion : Astragalus Polysaccharides can inhibit proliferation of gastric carcinoma cells, and arresting cell cycle of G0/ G1 phase may be one of the mechanisms.
