摘要
将筛选分离出的氯嘧磺隆降解菌2N3在特定的载体上扩大培养,进一步制备氯嘧磺隆降解菌菌剂。通过高效液相色谱法进行定量分析,根据降解率的高低筛选菌剂中载体配比、接种菌液量、加入营养液量、发酵时间、烘干温度等影响因子的单一因素最佳用量。设计5因素4水平正交试验,筛选出制备菌剂的最佳条件为:麦麸、木屑、玉米粉、稳定剂的最佳载体配比80∶10∶5∶5,接菌量15 mL,营养液用量10mL,发酵时间36 h,烘干温度35℃。上述条件下制备的菌剂对氯嘧磺隆的降解率为85.3%。对该菌剂的稳定性试验测得,在25℃下该菌剂能够稳定保存40 d,并且数天后仍对氯嘧磺隆具有降解作用。
This study screened and separated chlorimuron-ethyl-degrading bacterium 2N3 and cultivated it on specific carrier for further preparation of the microorganism bacterium degradation agent. Quantitative analysis was done by the method of high performance liquid chromatography where the degradation rate was the criteria to identify the optimum conditions of preparation factors such as carrier composition, amount of vaccination bacterium fluid, amount of adding nutrient solution, fermentation time and drying temperature etc. Carrier consisted of 80% wheat bran, 10% sawdust, 5% maize flour and 5% stabilizer was found to be the optimum recipe. The optimum conditions of individual factors included 15 mL of vaccination bacterium fluid, 10 mL of nutrient solution addition and 36 hours of fermentation time. The best drying temperature was 35℃. According to the single factor experiment, an orthogonal experiment consisted of five factors and four levels was designed and carried out. Through the comprehensive analysis of the factors, the optimum combination was as follows: Carrier with 80% wheat bran, 10% sawdust, 5% maize flour and 5% stabilizer; 15 mL of vaccination bacterium fluid; 10 mL of nutrient solution addition; 36 hours of fermentation and 30℃ of drying temperature. The degradation rate of chlorimuron-ethyl-degrading bacterium which was cultivated under mentioned conditions was 85.3%. Finally, from the stability test of aforementioned microorganism bacterium degradation agent, we can conclude that it can preserve good stability for 40 days at 25℃. In addition, the agent still exhibited degradation function after several days. The valuable significance of this study has provided experimental data and technical support for the commercial production of chlorimuron-ethyl-degrading bacterium 2N3.
