摘要
目的探讨SD大鼠缺氧缺血过程中脑组织水通道蛋白-4(aquaporin-4,AQP-4)和AQP-4mRNA的表达情况及意义。方法健康雄性SD大鼠60只,分为缺氧缺血性脑损伤(HIBD)组和对照组。HIBD组经右侧颈总动脉结扎后建立模型,对照组仅行假手术,不结扎颈总动脉缺氧。HIBD组和对照组于模型制成后0 h、6 h、1 d、2 d和3 d分别处死动物,对两组动物脑组织进行病理学观察,并检测脑组织中AQP-4蛋白和AQP-4mRNA表达。结果 HIBD组在模型建立后6 h、1 d、2 d和3 d均出现脑水肿并发生组织病学改变,随时间延长病理改变加重。与对照组比较,HIBD组脑组织AQP-4蛋白和AQP-4mRNA表达从HIBD后6 h即开始下降,2 d内下降至最低,3 d出现恢复。结论 AQP-4可能参与了大鼠脑内渗透平衡的维持,AQP-4表达下降可能与HIBD脑组织水肿和病理学变化的发生发展相关。
Objective To study the expressions of AQP-4 protein and AQP-4mRNA in brain tissue for SD rats with hypoxic-ischemic brain damage (HIBD), and discuss the significance. Methods 60 SD rats were divided into HIBD group and control group. HIBD group were subjected to the ligation of right carotid artery (ischemia) ,then they were put into a box full for an hour (hypoxia). The SD rats with HIBD model were divided into 5 groups (n =6) ,including 0 h,6 h,1 d,2 d and 3 d after establishment of HIBD. The normal control groups were also divided into 5 groups the same as HIBD groups. The Western Blot was used to detect the AQP-4 protein expression and RT-PCR was used to detect the AQP-4 mRNA expression in the brain tissue of both HIBD and the control groups. Results The brain edema and in- farction was present pathologically in brain tissues in HIBD groups after 6 h, 1 d,2 d and 3 d of HIBD and become more severe with time go- ing compared with that in the control groups. The expressions of AQP-4 protein and mRNA were significantly decreased in HIBD groups dur- ing the period of 6 h to 3 d compared with the control groups in the SD brain tissues. Conclusion The results show that the decreased ex- pression of AQP-4 is associated with brain edema and brain dmnage in SD rats with HIBD, and AQP-4 may play an important role in such brain damage conditions.
