龙葵碱诱导胰腺癌细胞Panc-1凋亡的实验研究 被引量:5

摘要 目的探讨龙葵碱对胰腺癌细胞Panc-1的凋亡诱导作用。方法采用经典方法培养胰腺癌细胞系Panc-1成功后,取对数生长期的细胞用于实验研究,用不同浓度(分别为20、30、40和50μg/mL)的龙葵碱进行干预,采用倒置显微镜观察龙葵碱作用后Panc-1细胞的形态学改变;CCK-8比色法检测龙葵碱对Panc-1细胞增殖的抑制作用;用流式细胞术AnnexinⅤ-FITC/PI双染法检测细胞凋亡率。结果龙葵碱显著抑制人胰腺癌细胞Panc-1生长,并具有浓度和时间依赖性。倒置显微镜观察发现,随着龙葵碱浓度增加和作用时间延长,Panc-1细胞表现出典型的凋亡形态改变;CCK-8比色法发现,随着龙葵碱浓度增加和作用时间延长,对Panc-1细胞的增殖抑制作用逐渐增强;流式细胞术检测发现,Panc-1细胞的凋亡率随着龙葵碱浓度增加而逐渐增加。结论①龙葵碱能够抑制胰腺癌细胞Panc-1的增殖,且此抑制作用具有时间和浓度依赖性。②龙葵碱具有诱导胰腺癌细胞Panc-1凋亡的作用,并具有浓度依赖性。 Objective To investigate the induction of apoptosis in human pancreatic cancer cell Panc-1 by Solanine.Methods Solanine of various available concentrations(20,30,40,50 μg/mL) were added to Panc-1 cell which stayed in logarithmic phase for the purpose of inhibiting the growth of Panc-1 cells.The morphology of cells was observed by inverted microscope;the inhibitory effect was detected with CCK-8 colorimetric method;and the cells apoptosis rate was detected with flow cytometry in Annexin V-FITC/PI double staining method.Results Solanine effectively inhibited the growth of Panc-1 in a manner of concentration-time dependent.Panc-1 presented typical apoptotic morpho-logical changes with the increase of drug concentration and response time under the observation of inverted microscope.And the inhibitory effect for the proliferation of Panc-1 gradually enhanced in a time-concentration dependent manner.Meanwhile,the apoptosis rate graduallyincreased with the increasingof drug concentration.Conclusion Solanine inhibited the proliferation of pancreatic cancer cells in a time and concentration dependent manner.Furthermore,the inhibitory effect may be achieved by the apoptosis induction effect of Solanine in a manner of concentration dependent.
