上海地区汉族人群中性粒细胞抗原的基因频率与其分子特征研究 被引量:5

Gene frequencies and molecular characteristic analysis of human neutrophil antigens in Shanghai Han population

摘要 目的:了解上海地区汉族人群人类中性粒细胞抗原(Human neutrophil antigens,HNA)的基因频率分布与其分子特征。方法:使用序列特异性引物聚合酶链反应(Polymerase chain reaction-sequence specific primer,PCR-SSP)对上海地区326名随机健康献血者的HNA-1(-a,-b,-c),HNA-3(-a,-b),HNA-4a(+,-)和HNA-5a(+,-)进行基因分型,测序和PCR-限制性片段长度多态性(PCR-restriction fragment length polymorphism,PCR-RFLP)验证其分子特征;并使用流式细胞术分析另外91份随机健康样本HNA-2a抗原表达。结果:本研究成功分析HNA-1、3-5各系统基因型。发现3例FCGR3B的变异体,其中2例为FCGR3B*01等位基因发生227 A→G和349 G→A的突变,另1例为一条染色体上的FCGR3B*02等位基因发生277 A→G的突变。HNA-1a、1b、1c、3a、3b基因频率分别为0.620、0.380、0、0.653和0.347,HNA-4a(+)、4a(-)、5a(+)、5a(-)基因频率分别为1、0、0.896和0.104,流式结果证实HNA-2a抗原频率为1,所测个体均具有HNA-2a阳性粒细胞,但不同个体具有不同比例的阳性粒细胞。这些分布与高加索人群相比有非常显著的差异,而与广东人群相比,主要是HNA-3系统分布有差异。结论:中国上海人群HNA具有独特的多态性,需要引起输血实践的注意。 Objective:This study aimed to know gene frequencies of human neutrophil antigens(HNA) and their molecular characteristic in Shanghai Han population.Methods:Polymerase chain reaction-sequence specific primer(PCR-SSP) amplification method was applied to HNA-1(-a,-b,-c),HNA-3(-a,-b),HNA-4a(+,-) and HNA-5a(+,-) genotyping in 326 unrelated healthy donors from Shanghai.HNA molecular characteristic was checked by PCR-based sequencing and PCR-restriction fragment length polymorphism(PCR-RFLP).HNA-2a expression was analyzed by flow cytometry for other 91 random healthy donors from Shanghai.Results:We analyzed the genotypes of HNA-1 and 3-5 systems successfully,and three FCGR3B variants was detected in this study,two of them were with 227 A→G and 349 G→A in FCGR3B*01 allele,while the other one with 277 A→G in FCGR3B*02 allele.The gene frequencies of HNA-1a,1b,1c,3a,3b were 0.620,0.380,0,0.653 and 0.347 respectively,and the gene frequencies of HNA-4a(+),4a(-),5a(+),5a(-) were 1,0,0.896 and 0.104 respectively.The antigen frequency of HNA-2a was 1,all individuals had HNA-2a positive neutrophils by flow cytometry,but different individual showed different proportion of HNA-2a positive neutrophils.HNA gene frequencies in our study were significant different from those in Caucasian in all HNA systems,while only frequencies of HNA-3 system were different from those in Guangzhou population.Conclusion:The polymorphism of HNA distribution in Shanghai population is unique,it deserves further attention in blood transfusion practice.
