介导重度TRALI的人类中性粒细胞抗原3(HNA-3)在不同种族人群中基因频率分布的研究 被引量:1

摘要 目的探讨编码人类中性粒细胞抗原-3(Human neutrophil antigens-3,HNA-3)的SLC44A2基因在江苏省汉族人群、美国白种人及美国黑人中的多态性及频率分布。方法采用序列特异性引物聚合酶链反应(Polymerase chain reaction-sequence specific priming,PCR-SSP)方法,分别对随机无亲缘关系的3个种族献血者的SLC44A2基因进行检测,其中包括江苏省的203名汉族、美国的102名白种人和245名黑人;同时采用PCR方法扩增相关外显子及侧翼内含子并进行测序分析,进一步验证PCR-SSP结果的准确性。结果汉族人群中HNA-3a/3a有91例,HNA-3a/3b 86例,HNA-3b/3b 26例;美国白种人中HNA-3a/3a有73例,HNA-3a/3b有26例,HNA-3b/3b有3例;美国黑人中HNA-3a/3a有212例,HNA-3a/3b有31例,HNA-3b/3b有2例。HNA-3a在江苏汉族人群,美国白种人及黑人中的基因频率分别为0.66,0.84和0.93;HNA-3b在江苏汉族人群,美国白种人及黑人中的基因频率分别为0.34,0.16和0.07。HNA-3b/3b纯合子在江苏省汉族人群的频率明显高于美国白种人及美国黑人,具有统计学意义(P<0.01)。结论编码HNA-3抗原的等位基因频率在3种不同种族中的分布存在多态性;该抗原的多态性及可能造成的TRALI需要引起临床上更多的关注。
