摘要
目的:动态观察脑卒中后大鼠行为学改变及颐脑解郁方对其早期的干预作用。方法:经Open-Field行为学评分后,将雄性Wistar大鼠随机分为正常组、假手术组、MCI组、中药组、西药组、PSD组、VD组。模型制备采用同种系微栓子体外注入法,由颈外动脉推注0.3ml同种异体血栓栓子;假手术组推注同等剂量生理盐水,并灌胃蒸馏水,中药组及西药组在模型制备后,予颐脑解郁方、尼莫地平干预给药4周;PSD组术后7d予21d慢性不可预知的温和应激加孤养复制PSD模型;VD组术后28d经水迷宫筛选后进入实验。模型复制成功后,分别于第1、2、4周进行穿梭箱主动回避反应实验,记录主动逃避(AAR)的次数、被动逃避(PAR)的次数以及被电击时间,并统计分析。结果:VD组的AAR、PAR次数最低,中药组的AAR、PAR次数均比未治疗组高,两组比较AAR及PAR都有显著性差异(P<0.05,P<0.01)。结论:颐脑解郁方有改善学习和记忆的功能,有利于脑卒中大鼠认知功能的恢复。
Objective: To observe the dynamic behavior changes and early intervention effects of Yi' nao Jieyu Formula af- ter stroke in rats. Methods: Scoring by the Open-Field behavior in male Wistar rats were randomly divided into normal group, sham-operation group, MCI group, treatment group, control group, PSD group and VD group. Model was prepared by in vitro injection of the same germ-line microemboli from the external carotid artery of 0. 3ml of allogeneic clot; sham operation group were injected the same dose of saline and intragastrically administrated with distilled water, Traditional Chinese Medicine group and western group were respectively administrated of four weeks with Yinao Jieyu Formula and Nimodipine after model prepared ; PSD model was prepared by chronic unpredictable mild stress plus solitary housed from 7d to 21d after operation; VD group were collected by the water maze filtered on 28th day after operation. Shuttle box active avoidance response experiment was respectively done on the 1Bt, 2"d and 4th week after success of model copying, and recording the initiative to escape (AAR) , the number of passive avoidance (PAR), the number of electric shock time, and statistically analyzing. Results: The frequency of PAR and AAR were the lowest in VD group, the frequency of PAR and AAR of Chinese medicine group were lower than the sham-opera- tion groups, the frequency of AAR and PAR had significant differences (P〈0. 05, P〈O. 01 ) between the two groups. Conclu- sion: Yi ' nao Jieyu Formula can improve learning and memory function, and it' s benefit to recovery of stroke cognitive func- tion in rats.
