摘要
[目的]观察和分析健脾温肾法对免疫抑制脾虚型小鼠免疫功能的调节作用及其机制,为临床应用提供实验研究依据。[方法]将ICR小鼠随机分为空白对照组、模型组和给药组,采用80mg.kg-1环磷酰胺诱(cydophosphamide,CTX)导免疫抑制的小鼠模型,同时给药组于造模第1d起予中药灌胃给药,连续10d。检测外周血白细胞计数、T淋巴细胞亚群分布及NK细胞。实验结束后解剖称重脾脏、胸腺,计算脾脏、胸腺指数。[结果](1)健脾温肾法能拮抗CTX对NK细胞的抑制作用;(2)健脾温肾法能拮抗CTX对白细胞的抑制作用。(3)健脾温肾法抑制CTX导致的小鼠代偿性脾肿大。(4)CTX对非T淋巴细胞有明显抑制作用,健脾温肾法对T淋巴细胞无明显作用。[结论]健脾温肾法对环磷酰胺免疫抑制型小鼠的免疫功能有增强作用。
[Objective] In order to explore the immunomodulating effects of warming the kidney and strengthening spleen on immuno deficiency mice.The study can provide experimental data for the clinical application of the treatment on patients with immuno deficiency.[Methods] ICR mice were randomly divided into control group,model group and treatment group.The models were injected by 80mg/kg cyclophosphamide into abdominal cavity to establish the immune suppressing model,meanwhile the treatment group were fed with Jiaweisijunzi tang for 10 days,the blood was taken from the mice filled with heparin,which was used to detect distribution of lymphocyte subsets and number of white blood cells.On the last day of the experiment,all the mice were put to death by taking off the neck,while their spleen and thymus were used to calculate the spleen and thymus index.[Results] 1.It prompted that the method of warming the kidney and strengthening spleen could antagonist the effect of the immunosuppression on NK cells.2.It prompted that it could antagonist the effect of the immunosuppression on WBC.3.It prompted that it had no effect on organs index.4.It prompted that CTX may have the effect of the immunosuppression on non-T lymphocytes,but it had no effect on T lymphocytes.[Conclusion]This method could enhance immune function of spleen deficiency and immune suppressing mice with immune adjustment effect.
