添加生长激素肠外营养对短肠大鼠代谢效应的影响 被引量:6

摘要 目的 研究单纯肠外营养 (PN)与添加生长激素 (GH)的PN对短肠大鼠的代谢作用。方法 建立PN短肠大鼠模型 ,随机分成普通肠外营养组 (STD组 )及添加GH的肠外营养组 (rhGH组 )。测各组大鼠每日体重及氮平衡 ,计算其累积氮平衡。实验结束后测各组织器官重量变化 ,并测大鼠总体水、脂肪及蛋白含量。结果 PN结束后rhGH组大鼠体重、每日氮平衡及累积氮平衡均显著高于STD组 ,P <0 .0 1。各器官重量分析显示rhGH组大鼠腓肠肌重量显著高于STD组 ,P <0 .0 5 ,但与对照组无明显差异。机体组成分析表明rhGH组大鼠总体蛋白及所占机体百分比较STD组显著升高 ,而总体脂肪则明显下降 ,P <0 .0 5。结论 PN时辅助添加rhGH具有显著的促代谢合成作用 ,蛋白合成增加是rhGH作用的显著特点。 Purpose To study the different metabolic effects of parenteral nutrition(PN) alone and PN combined with recombinant human growth hormone (rhGH) in parenterally fed,shortbowel rats. Methods Twenty SD rats were randomized divided into 2 groups:STD and rhGH group.Parenterally fed,short bowel rat models were established.The rats were weighted daily,nitrogen balance was calculated daily too and was used to calculate the accumulated nitrogen balance.After 6 days of PN,rats were killed,various organs were dissected and weighted,the carcasses were used for analysis of body composition. Results At the end of PN,rats in rhGH group weighed significantly more than STD grup,(193.4±15.78) vs ( 174.6± 14.62) g respectively.The average daily nitrogen balance and accumulated nitrogen balance in rhGH group was also higher than that in STD group.The 7 day accumulated NB were (1 252.9±294.3) vs (704.8±379.0) mg N/d respectively.The absolute weight of gastrocnemius muscle in rhGH group ( 2.68 ±0.34) g was significantly increased compared to STD group (2.18±0.17) g,but was not differnt from baseline group (2.55±0.28) g.Analyses of body carcass composition show that carcass weight and its percentage as protein was higher in rhGH group than in STD group,(21.1±3.2) vs (17.9±3.2) g and ( 19.5 ±2.8)% vs (16.4±2.4)% respectively,but carcass weight as fat was lower in rhGH group than in STD group,( 5.4 ±2.3) vs (8.5±3.9) g respectively. Conclusions rhGH significantly improve the anabolism in parenterally fed rats,the improvement of protein synthesis is characteristic of the rhGH effects.
