摘要
SAA抑制二甲苯诱发的小鼠耳肿或蛋清诱发的大鼠后足炎肿,使蛋清或酵母致大鼠后足炎症渗出液中PGE_2含量降低。SAA使蛋清诱导的小鼠炎足中的丙二醛生成减少;花生四烯酸则使之增加,此作用可被SAA完全取消,提示SAA抑制PG环加氧酶。SAA拮抗组胺(IC_(50)=89ug/ml).5—羧色胺(IC_(50)=38ug/ml)和过敏慢反应物质收缩豚鼠回肠的作用.对PGE_2收缩大鼠胃条的作用无明显影响。
The effects of total alkaloids of sophra alopecuroides L (SAA) on prostaglandin (PG) metabolism in inflammatory paws of rats and mice as well as its antagonistic action against inflammatory mediators were studied. The results showed that SAA inhibited rat hind paw swelling induced by egg white, leaving that by yeast unalterated; meanwhile, it decreased content of PGE_2 in inflammatory fluid from rat swelling paws irrited by both irritants. Content of malonyldialdehyde in mouse swelling paws stimulated by egg white was decreased by SAA and increased by arachidonic acid (AA), The action of AA could be entirely abolished by SAA, suggesting that PG cyclo-oxygenase was inhibited by SAA, Also, SAA directly antagonized action of 5-hydroxytrypatamine (IC__(50)=38μg/ml), histamine (IC_150)=89μg/ml), slow reaction substance of anaphylaxis (SRS-A), but the action of SAA was much weaker.It is concluded that SAA is a potent inhibitor of PG cyclo-oxygenase and antagonist of inflammatory mediators.
