Determination of Valproic Acid Concentration in Serum by Optimized HPLC after Pre-column Derivatization

摘要 目的:优化柱前衍生-高效液相色谱法监测血清中丙戊酸浓度的方法。方法:以乙腈为蛋白沉淀剂,以α-溴苯乙酮为衍生化试剂进行测定。色谱柱为ANAX丙戊酸专用分析柱,流动相为乙腈-水(70:30),柱温为40℃,流速为0.7mL·min-1,检测波长为266nm。结果:丙戊酸血药浓度在4.88~139.46μg·mL-1范围内线性关系良好(r=0.9996),平均回收率为100.01%,日内、日间RSD均<3%。结论:本法可简便、快速、稳定地测定血清中丙戊酸的浓度,能满足临床监测的要求。 OBJECTIVE: To optimize HPLC after pre-column derivatization to monitor serum concentrations of valproic acid. METHODS: The separation was performed on ANAX column with acetonitrile-water (70:30) as mobile phase at the flow rate of 0.7 mL·min-1 with detection wavelength of 266 nm using acetonitrile as protein precipitants and a-bromoacetophenone as derivating agent. RESULTS: The linear range of valproic acid was 4.88- 139.46 μg·mL-1(r= 0.999 6) with mean recovery of 100.01%. The RSDs of inter-day and intra-day were less than 3 %. CONCLUSION: This method is simple, rapid and stable for the determination of valproic acid concentration in serum, and it meets the demand of clinical monitoring.
