应用Con A,LCA交叉亲和免疫电泳探讨人血清AGP和α_1AT分子异构体

Studies on Molecular Variants of AGP and α_1AT in Human Sera by Con A and LCA Crossed Immuno-affino-electrophoresis

摘要 本文应用刀豆素A(Con A)和小扁豆凝集素(LCA)交叉亲和免疫电泳(CIAE)分离人血清α_1-酸性糖蛋白(AGP)及α_1-抗胰蛋白酶(α_1AT)分子异构体。结果表明:经 Con A-CIAE,正常人及胃癌患者 AGP 均显示三种异构体,孕妇、子宫肌瘤和卵巢癌患者 AGP 仅呈现二种异构体,且其中弱亲和性异构体含量显著减少。本结果初步提示,这些 AGP 异构体的变化可能与妊娠及生殖系统肿瘤有关。正常人及上述患者α_1AT 经 Con A-CIAE 可分离三种异构体,而经 LCA-CIAE 只分离二种异构体。AGP 与 LCA 无亲和性,表明 Con A 和 LCA 虽然具有相同的糖基结合专一性,但所识别的糖链构型可能不同。 Variants of α_1-acid glycoprotein(AGP)and α_1-antitrypsin(α_1AT) in human sera were separated by Con A and LCA crossed immuno-affino-electrophoresis(Con A-CIAE and LCA-CIAE).With Con A-CIAE,the serum AGP of normal human and that of patients with gastric cancer were all divided in to three variants,while that of pregnancy and patients with cancer of ovary and myoma of uterus showed only two variants and the amount of the weak-affino variants decreased.Results suggested that the changes of the AGP variants were probably correlated with pregnancy and the tumors of reproduc- tive system. Serum α_1AT also showed three variants by Con A-CIAE,while only two variants were disclosed by LCA-C1AE.No affinity were found between AGP and LCA.Thus,in spite of Con A and LCA specially bind the same glyeosyl residues,they perhaps recognize the different glyeosyl configurations.
