摘要
目的 解决在应用凝集素亲和免疫电泳自显影法检测甲胎蛋白异质体过程中对单一峰型的结果难以判断的问题。方法 对以往建立的凝集素亲和免疫电泳自显影法略作改进 ,在原来两块分别含凝集素和AFP抗血清的凝胶之间加一既不含凝集素也不含抗血清的凝胶条。选择 16例AFP异质体为均一型的血样本进行对照测定 ,并以凝集素亲和免疫电泳印迹法验证改良法的结果。结果 在只含凝集素的条件下表现为 1个峰的样本用改良法检测 ,良性肝病的样品峰型仍为 1个对称峰 ,恶性肝病的样品峰型由原来的 1个峰变为 2个峰。实验表明 ,应用改良法结果的重复性满意 ,并得到凝集素亲和免疫电泳印迹法的验证。结论 改良法使原本难以判断的结果清晰易辨 。
Objective To identify a single precipitin peak appeared in lectin-affinity immunoelectrophoresis autoradiography for the measurement of α-fetoprotein (AFP) variants. Methods The crossed immunoaffino-electrophoresis in the presence of lectin was modified. Briefly, an additional gel slab without either lectin or antibody was placed between the two original agarose gels, which also contain a sample well for the first dimension electrophoresis. 16 serum samples with a single precipitin peak of AFP by the unmodified method were simultaneously assayed using the modified method, and the results were confirmed by lectin-affinity electrophoresis or antibody-affinity blotting. Results Two possibilities were observed using the modified method: a single peak remained or turned into two peaks, which suggest a benign liver disease or a malignant liver disease respectively. The modified method has a good reproducibility, and the results accord with that by lectin-affinity electrophoresis and antibody-affinity blotting. Conclusion The modified method can differentiate lectin-reactive AFP and lectin-nonreactive AFP, and can be introduced in clinical use.
