摘要
目的 观察血糖变化对 STZ糖尿病大鼠骨骼肌葡萄糖转运体 (GL UT4)基因表达的影响。方法 用较小剂量的 STZ获得正常空腹血糖和空腹胰岛素水平的糖尿病大鼠 ,使用根皮苷纠正这种糖尿病大鼠进食后 2小时的高血糖 ,利用 Northern blot测定骨骼肌中 GL UT4m RNA的含量。结果 糖尿病大鼠的骨骼肌中 GL UT4m RNA的含量明显低于正常大鼠 (P<0 .0 1) ,根皮苷治疗在不影响体重和胰岛素水平的基础上 ,使糖尿病大鼠的血糖降低、骨骼肌中GL UT4m RNA的含量增加 (P<0 .0 1) ;根皮苷治疗对正常大鼠的血糖、胰岛素和 GL UT4m RNA无影响。结论 血糖变化参与调节糖尿病大鼠骨骼肌中 GL
Objective To investigate the effect of glycaemia on expression of GLUT4 in diabetic rat skeletal muscle.Methods A lower dose streptozotocin (55mg/kg·body wt) was used to establish diabetic models with normal fasting blood glucose and fasting serum insulin levels.Phlorizin was used to correct the 2 hours postprandial hyperglycaemia of the diabetic rats.The GLUT4 mRNA in skeletal muscle was assessed with Northern blot analysis.Results The diabetic rats had the same fasting blood glucose and serum insulin levels as the control,but much higher 2 hours postpra control.The GLUT4 mRNA in skeletal muscle of diabetics was significantly lower than that of controls ( P <0 01).Phlorizin injection partly corrected 2 hours postprandial hyperglycaemia and increased the GLUT4 mRNA content in skeletal muscle of diabetic rats ( P <0 01) without altering body weight gain and serum insulin levels.Phlorizin treatment had not effects on the fasting or 2 hours postprandial blood glucose,serum insulin levels and GLUT4 mRNA contents of skeletal muscles in controls.Conclusion These results indicated that glycaemia can regulate the GLUT4 expression in skeletal muscle of diabetic rat.
