SP600125对高血压全脑缺血再灌注大鼠学习记忆的影响

摘要 目的:探讨SP600125对高血压大鼠脑缺血再灌注损伤后学习记忆的影响。方法:雄性WKY大鼠随机分为假手术组和全脑缺血再灌注组(IR组);另取雄性自发性高血压大鼠随机分为高血压全脑缺血再灌注组和SP600125干预组。改良的Pulsineli4血管阻断(4-VO)法制作全脑缺血再灌注模型。脑缺血24、48h,电镜和光镜观察海马区神经细胞形态变化;免疫组织化学法检测海马区磷酸化JNK表达;八臂迷宫法测试动物学习记忆功能。结果:与假手术组比较,全脑缺血再灌注组海马区神经元结构损伤明显,神经元细胞存活密度降低;磷酸化JNK表达增高;学习记忆功能降低。与全脑缺血再灌注组比较,高血压全脑缺血再灌注组中海马区神经元结构损伤加重,神经元细胞存活密度降低;磷酸化JNK表达进一步增高;学习记忆功能降低。与高血压全脑缺血再灌注组比较,SP600125干预组海马区神经元结构损伤减轻;神经元细胞存活密度提高;磷酸化JNK表达进一步降低;学习记忆功能提高。结论:SP600125通过抑制脑组织神经细胞的丢失,改善高血压大鼠脑缺血再灌注损伤后学习记忆功能损伤。 Objective: To study the effects of SP600125 on learning and memory function after ischemia-reperfusion injury in hypertensive rats. Methods: Male WKY rats were divided into a sham group and a global cerebral ischemia group (IR group); and male spontaneously hypertensive wistar rats were divided into a hypertension with global cerebral ischemia group and a SP600125 treatment group. A global cerebral ischemia model was formed by improved four-vessel occlusion as described according to the description of Pulsinelli's method. Morphological changes in hippocampal region were observed under an electron microscope and a light microscope; The phosphorylated JNK expression was detected by immunohisto- chemistry; Learning and memory functions were evaluated with eight-arm maze. Results: Compared with the sham group, the neuronal morphous was damaged and the density of survival neurons was decreased; the phosphorylated JNK expression increased while the neuroscores of learning-memory function were decreased in the global cerebral ischemia group. Compared with the global cerebral ischemia group, the neuronal morphous was damaged and the density of survival neurons was decreased obviously~ the phos- phorylated JNK expression was increased and the neuroscores of learning-memory function were decreased significantly in hyper- tension with the global cerebral ischemia group. Compared with the hypertension with global cerebral ischemia group, the density of survival neurons was increased and the phosphorylated JNK expression was decreased, while the neuroscores of learning- memory function was increased in the SP600125 treatment group. Conclusion: SP600125 attenuates phosphorylated JNK activation, and can improve learning and memory function after global cerebral ischemia in hypertensive rats.
