摘要
目的:建立高效液相色谱/串联质谱法测定四环素软膏中四环素的含量。方法:采用Agilent SB-C18色谱柱(50 mm×2.1 mm,1.8μm),流动相为乙腈-0.3%甲酸溶液(10∶90),流速:0.2 ml.min-1。采用三重四级杆质谱检测器,正离子多反应监测模式(MRM),监测离子分别为:m/z 445/410,445/427。结果:四环素在0.80～4.00μg.ml-1范围内与峰面积呈良好的线性关系(r=0.999 1),平均加样回收率为99.3%,RSD=0.28%(n=6)。结论:本法简便易行,结果准确,重现性好,可用于四环素软膏中四环素的含量测定。
Objective： To establish a method for determining tetracyclini in tetracyclini ointments by LC-MS/MS.Method： An Agilent SB-C18（50 mm×2.1 mm,1.8 μm） column was used with acetonitrile-0.3% methanoic acid（10∶ 90） as the mobile phase,and the flow rate was 0.2 ml·min-1.MS/MS detection in multiple reactions monitoring（MRM） mode using positive electrospray ionization as the ionization source was applied to determine positive ions at（m/z） 445/ 410 and 445/427.Result： Tetracyclini was linear within the range of 0.80~4.00 μg·ml-1（r=0.999 1）,the average recovery was 99.3% and RSD was 0.28%（n=6）.Conclusion： The method is simple,rapid,accurate and reliable,and can be used in the determination of tetracyclini in tetracyclini ointments
