链脲佐菌素诱导1型糖尿病大鼠十二指肠磷酸糖蛋白的表达

摘要 目的观察链脲佐菌素(STZ)诱导的1型糖尿病大鼠早期十二指肠中磷酸糖蛋白(P-glycoprotein,P-gp)的变化,探讨P-gp在糖尿病中的重要作用。方法 SD雄性大鼠20只随机分为正常组及糖尿病组,用STZ复制1型糖尿病动物模型,饲养7 d后测血糖及尿糖,6周后进行灌注固定,利用免疫组织化学方法观察糖尿病组和正常组大鼠十二指肠绒毛高度及密度的改变,并分析P-gp的表达情况。结果糖尿病组大鼠的十二指肠绒毛高度显著增长(P<0.05),绒毛密度变化不明显,上皮细胞P-gp的表达增强(P<0.05)。结论糖尿病大鼠十二指肠吸收面积增加,P-gp的高表达可能导致耐药性的产生。 【Objectives】 To observe the expression of P-gp on the duodenum in type I diabetic rats,in order to study the influence of P-gp in DM.【Methods】 20 male SD rats were randomly divided into two groups: 10 normal control rats and 10 rats induced with STZ DM.7 days later,blood glucose and GLU were measured,6 weeks later,the immunohistochemical methods was used to detect changes of the duodenal naps height,density and P-gp positive area on the duodenum.【Results】 The height and P-gp positive area of duodenal nap in the DM group was higher than those in the normal group(P 0.05),but the density wasn't altered.【Conclusion】 The DM rats' duodenal absorption area was increased.The high expression of P-gp may lead to drug resistance.
