摘要
运用Fura 2显微荧光测量技术 ,在单个大鼠肾上腺嗜铬细胞上 ,测量中药知柏地黄丸浸液对胞内游离钙浓度 ([Ca2 + ]i)的作用 .实验发现 ,知柏地黄丸浸液作用于大鼠嗜铬细胞 ,数s内即可导致 [Ca2 + ]i的升高 ;而将标准胞外液换为无钙胞外液或加入L 型钙通道抑制剂均可抑制知柏地黄丸浸液对 [Ca2 + ]i的作用 .这表明知柏地黄丸浸液可以引起细胞外Ca2 + 内流 ,而该内流主要通过L 型钙通道的开放 .
By using microflurescent calcium measurement technique, the effects of extract of Zhi Bo Di Huang Wan on the intracellular calcium concentration ([Ca 2+ ] i) of single rat Chromaffin cell were studied. The results showed the extract caused an increase of [Ca 2+ ] i when it was puffing to the cell. Removing Ca 2+ from the bath solution or nifedipine obviously inhibited the increasement of [Ca 2+ ] i induced by the extract. It illustrated that the dependence of the Zhi Bo Di Huang triggered calcium signal on extracellular Ca 2+ and the opening of L Ca 2+ Channel made a great contribution to the [Ca 2+ ] i increase.
