因宁片对甲亢性肝损害模型大鼠肝脏细胞色素P_(450)基因表达的影响 被引量:1

Effects of Yinning Tablet on Gene Expression of Cytochrome P 450 in Liver Tissue of Rats with Hyperthyroidism Liver Damage

摘要 目的:研究因宁片对甲亢性肝损害模型大鼠肝脏细胞色素P450(CYP450)基因表达的影响。方法:采用L-甲状腺激素钠灌胃以复制甲亢性肝损害大鼠模型。实验分为6组,即空白对照、模型、甲亢灵和因宁片高、中、低剂量(2.4、1.2、0.6g.kg-1)组。通过实时荧光定量聚合酶联反应(PCR)方法检测因宁片对肝组织内CYP1A2、CYP2E1和CYP3A2基因表达的影响。结果:与模型组比较,因宁片高、中剂量组大鼠肝脏CYP2E1的表达显著减弱(P<0.05),CYP1A2和CYP3A2的表达无明显改变(P>0.05)。结论:因宁片能下调甲亢大鼠肝脏CYP2E1基因的表达,这可能是因宁片治疗甲亢性肝损害的分子作用机制之一。 OBJECTIVE:To study the effects of Yinning tablet on gene expression of cytochrome P 450(CYP 450)in liver tissue of rats with hyperthyroidism liver damage.METHODS:Rats were perfused with levothyroxine sodium intragastrically to establish the rat model of hyperthyroidism liver damage.The model rats were divided into 6 groups,i.e.blank control group,model group,Jiakangning group and Yinning tablet high-dose,medium-dose and low-dose groups(2.4,1.2,0.6 g.kg-1).The expressions of CYP1A2,CYP2E1 and CYP3A2 in liver tissue were detected by real-time quantitative PCR.RESULTS:Compared with model group,the expressions of CYP2E1 was decreased significantly(P〈0.05),and the expressions of CYP1A2 and CYP3A2 had no significant change(P〉0.05).CONCLUSION:Yinning tablet may down-regulate the expression of CYP2E1 in liver tissue of rats with hyperthyroidism liver damage,which may be one of the molecular action mechanisms that Yinning tablet prevent hyperthyroidism liver damage.
