摘要
目的检测胃癌组织中S100A2基因的mRNA和蛋白表达水平,分析S100A2基因在胃癌发生、发展过程中的作用机制。方法应用免疫组化、Western Blotting技术、RT-PCR技术检测胃癌组织中S100A2mRNA、蛋白表达水平的改变。结果 S100A2蛋白定位于胃黏膜上皮细胞的胞浆及胞核,胃癌组织中S100A2mRNA、蛋白表达下调,低分化胃癌与淋巴结转移胃癌S100A2表达降低最明显。结论胃癌组织S100A2蛋白、mRNA表达降低,且其表达与胃癌分化程度和淋巴结转移相关。
【Objective】To evaluate the possible role mechanism of S100A2 gene in the process of carcinogenesis and development in stomach,the levels of mRNA and protein of S100A2 gene were detected.【Methods】The expression state of mRNA and protein of S100A2 gene were analyzed by RT-PCR,S-P immunohistochemical assay and Western blotting respectively in gastric carcinoma.【Results】The S100A2 staining was located on the nuclear membrane and in the cytoplasm.The levels of S100A2 mRNA and protein in gastric carcinoma were significantly lower than that in normal gastric mucosa,and the expression of S100A2 gene was the lowest in the poorly differentiated gastric carcinoma and gastric carcinoma with lymph metastasis.【Conclusions】The expression of S100A2 gene,which was correlated with differentiation degree of gastric carcinoma and lymph metastasis,was significantly lower in gastric carcinoma than that in normal gastric mucosa.
