摘要
目的通过研究溃疡性结肠炎(UC)模型小鼠结肠组织中TLR2、TLR3、TLR5和TLR9的表达随时间变化的关系,探讨Toll样受体家族在UC致病机制中的作用。方法 60只BALB/c小鼠随机分为3组,第1组不予处理,第2组予5%葡聚糖硫酸钠(DSS)溶液自由饮用7 d造模,第3组予5%DSS溶液自由饮用14 d造模。取各组小鼠的结肠标本,用RT-PCR和Western blotting的方法半定量检测TLR2、TLR3、TLR5、TLR9的表达。结果①正常小鼠结肠中无TLR2、TLR3、TLR5和TLR9表达;②造模7 d小鼠结肠中无TLR3和TLR5表达,而TLR2和TLR9出现明显表达;③造模14 d小鼠结肠中仍然无TLR3和TLR5表达,而TLR2和TLR9的表达则继续增加。结论 TLR2和TLR9可能参与了溃疡性结肠炎的致病机制,而TLR3和TLR5可能未参与UC的致病机制。
【Objective】To explore the role of Toll-like receptors(TLRs) family in ulcerative colitis(UC) pathogenesis by investigating the change of Toll-like receptor 2(TLR2),TLR3,TLR5,TLR9 over time in colon of ulcerative colitis mice.【Methods】60 BALB/c mice were randomly divided into 3 groups,Group 1 was untreated,Group 2 was feed with 5% dextran sodium sulfate(DSS) for 7 days,Group 3 was feed with 5% DSS for 14 days.The level of mRNA and protein of TLR2,TLR3,TLR5,TLR9 in colon was detected using RT-PCR and Western blotting.【Results】There was no expression of TLR2,TLR3,TLR5,TLR9 in colon of normal mice.After 7 days induced with DSS,the expression of TLR2,TLR9 was showed detectable in the colon,and their expression was continued increasing after induction for 14 days.The expression of TLR3 and TLR5 was not detected in three groups.【Conclusions】These results indicated TLR2,TLR9 may be involved in the pathogenesis of UC,while no clue showed TLR3,TLR5 played any roles in the pathogenesis of UC.
