幽门螺杆菌对食管癌COXⅠ表达的影响 被引量:2

摘要 目的观察幽门螺杆菌(H.pylori,Hp)作用于食管癌细胞后细胞色素氧化酶Ⅰ(cytochrome oxidaseⅠ,COXⅠ)mRNA及其蛋白表达的变化,探讨Hp在肿瘤发生发展中可能的作用机制。方法将Hp与食管癌细胞分别作用4、8、24 h,收集细胞,应用Real-Time PCR和Western blot法检测各时间点COXⅠmRNA及其蛋白表达。结果 Hp与食管癌细胞共培养4、8、24 h后,COXⅠmRNA表达量逐渐减少,4 h的表达量显著低于对照组,其后下降速度减慢,24 h后表达量不再有明显改变。COXⅠ蛋白表达呈现相似的趋势。结论 Hp与食管癌细胞共培养后,可导致线粒体编码基因COXⅠmRNA及其蛋白表达障碍,从而影响线粒体功能。 Objective To study the effects of H. pylori (Hp) on the expressions of COX Ⅰ mRNA and protein in esophageal carcinoma cells, and explore the potential role of Hp in carcinogenesis. Methods After ECa-109 cells were treated by Hp for 4, 8, 24 h respectively, the expression of COX Ⅰ mRNA and protein was detected by Real-Time PCR and Western blot. Results After ECa-109 cells were treated by Hp for 4, 8, 24 h respectively, the expression of COX Ⅰ mRNA de- creased gradually, after exposure to Hp for 4 h, the expression of COX Ⅰ significantly lower than control group, than the decreasing speed slowed When treated with Hp for 24 h, the expression have no significant changes. The expression of COX Ⅰ protein revealed the same trend. Conclusion Hp can affect mitochondfial function by down regulating the expres- sion of mitochondrial encoding eytochrome oxidase Ⅰ gene.
