多维色谱-质谱联用技术分析胰腺癌血清差异表达蛋白 被引量:1

摘要 目的:探讨胰腺癌患者血清和健康人血清之间蛋白质的差异表达,寻找特异性的肿瘤生物标志物。方法:应用多维色谱-基质辅助激光解吸电离飞行时间质谱(MALDI-TOF MS)方法,检测23例胰腺癌患者和30例健康人血清蛋白质谱图表达。结果:与健康人血清比较,胰腺癌患者血清谱图中显示1个特异性蛋白质峰明显增高,相对分子质量为5734D,其特异性和敏感性高。结论:人胰腺癌血清特异性蛋白质的发现,对进一步研究肿瘤发生机理及其临床特异性生物标志物的检测等具有重要意义。 Objective:To explore differential expression of serum protein between patients with pancreatic carcinoma and healthy controls and to identify specific tumor biomarkers.Methods: The protein spectra of 23 samples from patients with pancreatic carcinoma and 30 samples from healthy controls serum were detected by multidimensional chromatography-MALDI(surfaced enhanced laser desorption/ionization) technology.Results: Compared with healthy people serum,one specific protein peak with molecular weight of 5734D relatively was significantly increased in patients with pancreatic carcinoma,and the specificity and sensitivity were relevantly increased.Conclusion: Detection of specific protein in human pancreatic carcinoma is significant both for study on cancer development mechanism and determination of clinical specific biomarkers.
