摘要
DNA结合蛋白的化学合成、结构与功能研究有助于阐明基因调控过程的分子机制。主要介绍反相高效液相色谱法(RP-HPLC)在化学合成DNA结合肽分析纯化中的应用。
The studies of their structure and function favour the understanding of molecular mechanism for gene regulation. Leucine -Zipper GCN-4(No.1), helix-turn-helix C-myb protein No. 2 and zinc-finger No.3 polypeptide were designed and synthesized. Their sequences are as following:No.1 DPAALKRARNTEAARRSRARKLQRMKQGGC No.2 Troc-TDDWKVIANYLPNRTDVQCQHRWQKVLNPE No.3 CRCMCNIEGHYANECP-NH2Separation detection using double wave length, analysis of non-enantiotropic polypeptides, monitor of reactive process and purification of polypeptides were carried out by means of revered-phase high performance liquid chromatography (RP-HPLC). The columns Cosmosil 5C-C_(18) AR (10 × 250mm) and YMC-gel ODS (20 ×250mm) with mobile phases 0. 1% TFA-H_2O and 0.1% TFA-CH_3CN are adopted selectively. The results show that the RP-HPLC is one of the chief methods of analysis and purification for protein and polypeptides with the advantages of high resolution, high speed, and easy application. Analysis and purification of DNA binding polypeptides have been used for studying of structure and function .
