摘要
目的:探讨白细胞介素1α(IL-1α)及其受体拮抗剂(IL-1ra)对大肠癌细胞株增殖、侵袭的影响。方法:选用高肝转移潜能的大肠癌细胞株WiDr及低肝转移潜能的大肠癌细胞株Caco-2和Colo-320,分别用RT-PCR和ELISA方法检测IL-1α及IL-1 I型受体(IL-1R I)的基因及蛋白在3种细胞中表达;用不同浓度的外源性IL-1α或IL-1ra作用于上述细胞后,用WST-1法检测3种细胞的增殖情况以及Transwell小室检测WiDr与Caco-2细胞侵袭能力。结果:RT-PCR显示IL-1α基因仅在高肝转移潜能的WiDr细胞中表达,在低肝转移潜能的Caco-2和Colo-320细胞中不表达,而IL-1R I基因在3种细胞中均有表达;ELISA检测WiDr细胞IL-1α蛋白浓度为(7.85±0.52)pg/mL,而Caco-2和Colo-320细胞未检测到;外源性IL-1α能明显增加Caco-2和Colo-320细胞的增殖能力(均P<0.05),对WiDr细胞的增殖无明显影响(P>0.05),但外源性IL-1ra能明显抑制WiDr细胞的增殖(P<0.05);外源性IL-1α能明显增强Caco-2细胞的侵袭能力(P<0.05),但对WiDr细胞侵袭能力无明显影响(P>0.05),外源性IL-1ra能明显抑制WiDr细胞侵袭(P<0.05),但对Caco-2细胞的侵袭能力无明显作用(P>0.05)。结论:大肠癌的肝转移潜能与IL-1α表达有关,外源性IL-1ra能抑制大肠癌细胞的增殖与侵袭。
Objective: To investigate the effects of interleukin 1α (IL-1α) and its receptor antagonist (IL-lra) on the proliferation and invasion of human colon cancer cells with different metastatic potentials as well as the role of IL-1α and IL-lra in metastatic process. Methods: The colonic carcinoma cell lines with high liver metastatic potential (WiDr) and low liver metastatic potential (Caco-2 and Colo-320) were used. The gene and protein expressions of IL-la and its type I receptor (IL-IR I) were determined by RT-PCR and ELSIA assay, respectively. After exposure to different concentrations of the exogenous IL-1α and IL-lra, the cell proliferation of the three cell lines was measured by WST-1 assay and the invasive ability of WiDr and Caco-2 cells was assessed by using TransweU chamber. Results: q-he RT-PCR results showed that the IL-1 a mRNA was only expressed in the WiDr cells of high liver metastatic potential but not in Caco-2 and Colo-320 cells of low liver metastatic potential, while the IL-1R I gene was expressed in all the three types of cells. The concentration of IL-la protein detected in WiDr cells by ELISA assay was (7.85+0.52) pg/mL but was not detected in Caco-2 and Colo-320 cells. The exogenous IL- Ia significantly increased the proliferative ability of Caco-2 and Colo-320 cells (all P〈0.05), but had no effect on that of WiDr cells (P〉O.05), while the exogenous IL-lra markedly inhibited the proliferation of WiDr cells (P〈0.05). The exogenous IL-la significantly enhanced the invasive ability of Caco-2 ceils (P〈O.O5), but had no influence on that of WiDr cells (P〉0.05); the exogenous IL-lra significantly inhibited the invasiveness of WiDr cells (P〈0.05), but had no effect on Caco-2 cells (P〉0.05). Conclusion: The liver metastatic potential of colon cancer is related to its IL-la expression, and exogenous IL-ira can inhibit the proliferative and invasive ability of colon cancer cells.
