摘要
为探讨缺氧缺血性脑病 (HIE)新生鼠中枢神经系统一氧化氮合成酶 (NOS)的变化 ,本文采用NADPH二氧硫辛酰胺脱氢酶组织化学方法观察其脑组织中NOS分布的改变 ,并采用同位素生化技术直接测定大脑皮层、海马、小脑等处NOS活性。结果显示 :急性缺氧时脑组织中NOS分布及NOS活性与正常组相比 ,差异无显著性 (P >0 .0 5 )。但在HIE组 ,大脑皮层、海马、小脑等处NOS阳性胞体明显增多 ,NOS活性明显增高 ,差异有极显著性意义 (P <0 .0 1) ,且脑血管内膜NOS阳性反应明显加深。提示一氧化氮在缺氧所致脑损伤及脑循环调节中起一定作用。
In order to study the changes of nitric oxide synthase (NOS) in central nervous system in neonatal rats with hypoxic-ischemic encephalopathy(HIE),the NADPH diaphorase histochemistry test was taken to show the distribution of NOS in brain tissue and isotopic biochemical assay was taken to determine the activity of NOS in cerebral cortex、hippocampus、cerebellum.The results showed that copmared with normal fats.the distribution and the activity of NOS in brain tissue had no significance in the acute hypoxic rats(P>0.05).However,in HIE neonatal rats,the numbers of NOS-positive neuronal bodies and the activity of NOS were markly increased in cerebral cortex、hippocampus、cerebelum(P<0.01) and the NOS-positive reaction products of brain vasculsar endothelium were markly enhanced, too.It suggests that nitric oxide plays partly a role in brain injury and circular regulation caused by hypoxia.
