摘要
以始兴石斛种子为外植体,研究了NAA、6-BA、IBA、蔗糖以及活性炭(AC)等对种子无菌萌发及快速繁殖的影响。结果表明:适合种子萌发的培养基是MS+6-BA1.5mg.L-1+NAA 0.6mg.L-1+AC0.1%,萌发率可达100%;适合原球茎增殖的培养基是MS+6-BA0.8mg.L-1+NAA0.3mg.L-1+蔗糖4.5%,增殖系数为7.75倍;适合分化的培养基是MS+6-BA2.5mg.L-1+NAA0.5mg.L-1+AC0.03%,分化率为92%;适合生根的培养基是1/2MS+IBA0.5mg.L-1+AC0.05%,生根率为95.3%;以苔藓和树皮作为基质,试管苗移栽成活率可达93%以上。
We studied the effects of the concentration and the ratio of NAA,6-BA,IBA,sucrose and active carbon on the germination of seeds and their rapid propagation of Dendrobium shixingense,by using its seeds as explants,.Our results indicated that the optimum medium for germination of seeds was MS containing 1.5 mg·L^-1 6-BA,0.6 mg·L^-1 NAA and 0.1% active carbon,with the germination rate being 100%.For multiplication of protocorms,the MS containing 0.8 mg·L^-1 6-BA,0.3 mg·L^-1 NAA and 4.5% sucrose was the suitable medium,and its propagation coefficient was 7.75.The Differentiation medium was best in the MS containing 2.5 mg·L^-1 6-BA,0.5 mg·L^-1 NAA and 0.03% active carbon,whose differential rate reached 92%.As for rooting,the 1/2 MS supplemented with 0.5 mg·L^-1 IBA and 0.05 active carbon was preferred,in which the rooting rate reached 95.3%.,The survival rate of the rooting plantlets transplant was more than 93% in the case of using the moss and bark as ground.
