摘要 建立了测定海参中羟脯氨酸(hydroxyproline,Hyp)含量的高效液相色谱法。样品经酸水解后,用4-二甲基胺基偶氮苯-4-磺酰氯(DABS-Cl)衍生,以乙腈-柠檬酸溶液为流动相梯度洗脱,经C18柱分离,436nm下紫外检测器检测。结果表明,羟脯氨酸衍生物与样品基质很好分离,在40~2000μg/L范围内线性关系良好(r>0.999),平均回收率84.2%~104%,RSD<10%。此法灵敏、准确,适用于海参中羟脯氨酸含量的测定。 The study is meant to determine the content of hydroxyproline in sea cucumber. After hydrolysis, the sample was decomposed into amino acids mixture, including hydroxyproline. The amino acids were derived with DABS - Cl. Liquid chromatographic separations were performed on a C18 column with a citric acid buffer - acetonitrile gradient elution program. UV detector was used to determine the hydroxyproline at 436 nm wavelength. The results indicated that hydroxyproline can be well separated from other substances. The linear range was from 40 - 2 000 μg·L -1( r 〉 0. 999). The recoveries of spiked samples were 84.2% - 104% , and the RSD was less than 10%. The proposed method is sensitive, accurate and suitable for the determination of hydroxyproline in sea cucumber.
