电针对神经干细胞海马内移植老年性痴呆大鼠行为学及海马超微结构的影响 被引量:4

Effect of Electric Needle on Metabolism of Memorizing Ability and on the Ultrastructure of Hippocampus

摘要 目的:观察电针对神经干细胞(NSCs)海马内移植老年性痴呆(AD)大鼠行为学及海马超微结构的影响,探讨电针治疗老年性痴呆的机制。方法:采用β-淀粉样蛋白(β-AP)向海马CA1区定向注射制做AD模型,将大鼠分为四组:正常组、模型组、移植组、移植电针组,每组各12只。针刺选取百会、大椎、人中三个穴位。大鼠跳台检测行为学指标;通过电镜观察海马组织超微结构。结果:移植电针组大鼠跳台潜伏期比模型组增加(P<0.05),错误次数比模型组减少(P<0.05);电镜观察到AD大鼠脑组织海马区神经元均有不同程度的破坏,模型组大鼠海马区神经元细胞质内线粒体明显肿胀、变性、嵴断裂;移植电针组大鼠海马区神经元细胞损伤程度比模型组明显减轻。结论:电针结合神经干细胞移植对于AD模型大鼠的神经元结构具有明显的改善作用,减少神经元的损伤。 Objective : To observe the effect of electric needle on the study of memorizing ability and the effect on the ultra- structure of hippocampus and discuss the mechanism of electric needle for curing AD. Methods:The model was made by injecting 13 -AP to CA1 of hippocampus. The rats were randomized into normat, model, transplantal and transplantal nee- dle group, each had 12 rats. The acupuncture was selected at baihui, dazhui and renzhong and connected the electronic needle device. The memorizing ability was measured by using the diving tower of rats and the ultrastructure of hippocam- pus was observed by electron microscope. Result : The transplantal needle incubation of step down increased ( P 〈 0. 05 ) than the model group and the correct times decreased (P 〈 0. 05 ) than the model ; the nerve cell of AD rats in brain hip- pocampus had different damages. The uhrastructure of mitochondria in the cytoplasm had been damaged including mito- chondria degeneration and cristae split by using electron microscope in the model ; while transplantal needle group clearly decreased than the model. Conclusion:The structure of nerve cell has been clearly improved by electric needle com- bined with transplant for AD rats.
