TaqMan探针分析p62/IGF2BP2基因多态性

摘要 目的在临床实践中建立一种有效的检测p62/IGF2BP2基因多态性的方法。方法盐析法提取40例肝癌患者的外周血基因组DNA。采用实时荧光PCR TaqMan探针方法对p62/IGF2BP2的一个单核苷酸多态性位点:rs11705701进行基因分型分析,并进行基因序列测序验证。结果通过对肝癌DNA进行测试分析,表明该位点在肝癌样品中存在多态性。结论目前的数据初步表明TaqMan探针方法能够用于IGF2BP2基因多态性分型分析。 Objective To set up a highly efficient method to detect the mutation of p62/IGF2BP2 in clinical practice. Methods The genomic DNAs were isolated from 40 hepatocellular carcinoma (HCC) patients' blood samples. The TaqMan real time PCR technique was used to detect the mutation of one of the single nucleotide polymorphism (SNP) : rs11705701 in p62/IGF2BP2 from the HCC DNA samples. Results There are SNP mutations in p62/IGF2BP2. Conclusions The TaqMan probe is a highly efficient, economical screening method to detect the SNP mutation of p62/IGF2BP2.
