原发性胆汁性肝硬化患者外周血T细胞microRNA表达谱变化 被引量:10

摘要 目的观察原发性胆汁性肝硬化(PBC)患者外周血T细胞中微小RNA(microRNA,miRNA)表达谱的变化。方法选取20例PBC患者,收集同期20份健康人外周血作对照。应用芯片技术筛选PBC患者外周血T细胞中表达差异的miRNA,并用实时定量PCR进行验证。结果与健康对照组相比,PBC患者外周血T细胞中有25个miRNA表达失调,其中有23个下调,仅有2个上调(均P〈0.05)。定量PCR显示,miR-346、miR-17-5p、miR-20a和miR-let-7b表达显著低于健康对照,而miR-451和miR-129表达显著高于健康对照,与芯片结果一致。结论PBC患者外周血T细胞miRNA表达与健康对照比较存在差异,这些异常表达的miRNA可能参与PBC的发病。 Objective To explore the expression pattern of microRNA (miRNA) in T cells of peripheral blood mononuclear cell (PBMC) from patients with primary biliary cirrhosis (PBC). Methods The expression profile of miRNA in T cells of PBMC was determined by microarray assay and validated by real-time quantitative reverse transcription-polymerase chain reaction (qRT-PCR). Results In comparison with the healthy controls, 23 miRNA were down-regulated and 2 miRNA had a higher expression ( all P 〈 0. 05). As revealed by qRT-PCR, the expressions of miR-346, miR-17-Sp, miR-20a and miR-let-Tb decreased obviously while miR-451 and miR-129 became up-regulated. The results were in agreement with those of microarray. Conclusions The PBG patients and healthy controls have significantly different expression profiles of microRNA in T cells of PBMC. The differential expression of microRNA may be involved in the pathogenesis of PBC.
