摘要
目的探讨环氧合酶-2(COX2)在慢性非细菌性前列腺炎(CNBP)导致的大鼠神经痛中的变化及意义。方法将大鼠随机分为0周、2周、4周、6周、8周、12周和12周对照组,采用免疫法建立CNBP神经痛模型。观察大鼠一般情况变化、前列腺病理组织学变化。采用免疫组化SABC法检测大鼠前列腺和脊髓后角COX2表达变化。结果与0周组相比,2、4、6、8、12周组前列腺和脊髓后角COX2均呈阳性表达(P〈0.05)。0周组和12周对照组前列腺和脊髓COX2表达差异均无统计学意义。与2周组相比,8和12周组前列腺COX2阳性例数增多(P〈0.05),6、8、12周组脊髓COX2阳性例数增多,差异具统计学意义(P〈0.05)。与4周组比,8、12周组前列腺COX2阳性例数增多,差异具统计学意义(P〈0.05)。与6周组比,8、12周组前列腺COX2阳性例数增多,差异具统计学意义(P〈0.05)。进一步用确切概率法统计分析,2、4、6、8、12周前列腺和脊髓与0周组比(COX2阳性表达增加(P〈0.05)。其余各组相比差异无统计学意义。COX2在前列腺和脊髓后角中呈正相关。结论提示COX2可能参与了CNBP炎导致的大鼠神经痛的形成和维持。
Objective Investigate the changes and effects of cyclooxygenase-2 in the development ofneuropathic pain induced by chronic nonbacterial prostatitis in rats. Methods The rats were assigned randomly into the 0-, 2-, 4-, 6-, 8-, 12-Week Group, and the 12-Week Control Group. A chronic nonbacterial prostatitis model was established by the immunization method. The expressions of COX2 in the prostate gland and the posterior horn of the lumbar segment of spinal cord were detected by the immunohistochemical SABC method. Results Compared with those in the 0-Week Group, there were statistically significant differences of the COX2 expressions in the prostate gland and the posterior horn of spinal cord in the 2-, 4-, 6-, 8-, and 12-Week Groups, respectively. Compared with those in the 2-,4-,6-Week Group, there were statistically differences of the COX2 expressions in the prostate gland in the 8- and 12-Week Groups; Compared with those in the 2-Week Group, there were statistically differences of the COX 2 expressions in the posterior horn of spinal cord in the6-, 8- and 12-Week Groups. The COX 2 expressions in the prostate gland and the posterior horn of spinal cord were positively correlated. Conclusion These data indicated that COX2 might be involved in development of neuropathic pain induced by chronic nonbacterial prostatitis.
