摘要
目的观察丹红软肝胶囊含药血清对肝星状细胞(HSC)增殖及分泌的相关细胞因子的干预作用。方法 15只SD大鼠按随机数字法分为3组,即丹红软肝胶囊组、复方鳖甲软肝片组和正常对照组,各5只,通过灌胃制备丹红软肝胶囊及复方鳖甲软肝片药物血清。将HSC-6细胞分为3组,分别加入含丹红软肝胶囊、复方鳖甲软肝片药物血清及正常小鼠血清的培养基,培养24 h后,采用噻唑蓝(MTT)法检测HSC的增殖率,收集培养上清,采用酶联免疫吸附法(ELISA)检测Ⅰ型胶原及转化生长因子β1(TGF-β1)的含量。结果丹红软肝胶囊组、复方鳖甲软肝片组吸光度(OD)值均低于正常血清对照组(P<0.01);丹红软肝胶囊组、复方鳖甲软肝片组HSC细胞增殖的抑制率比较差异无统计学意义(P>0.05)。丹红软肝胶囊组、复方鳖甲软肝片组培养上清中Ⅰ型胶原、TGF-β1的含量与正常血清对照组比较均降低(P<0.01);丹红软肝胶囊组、复方鳖甲软肝片组培养上清中Ⅰ型胶原、TGF-β1的含量比较差异无统计学意义(P>0.05)。结论丹红软肝胶囊可抑制活化的HSC产生TGF-β1,并对HSC分泌胶原产生抑制作用,进而抑制HSC的增殖,这可能是其抗纤维化作用机制之一。
Objective To study of the effect of blood serum supplemented with Danhong Rangan capsule on proliferation of hepatic stellate cells.Methods 15 SD rats were randomly divided into three groups,Danhong Rangan capsule group,Fufangbiejiaruanganpian group and normal control group.Blood serum was supplemented with Danhong Rangan capsule and Fufangbiejiaruanganpian drug by lavage.Rat hepatic stellate cells were divided into three groups： normal control group was added with normal serum,Danhong Rangan capsule group were added with blood serum supplemented with Danhong Rangan capsule compound,Fufangbiejiaruanganpian group were added with blood serum supplemented with Fufangbiejiaruanganpian compound.Cells in each group were cultured for 24 hours.MTT were applied to measure the HSC-T6 proliferation.The expressions of type Ⅰcollagen and TGF-β1 among each group were analyzed by ELISA.Results The absorbance in Danhong Rangan capsule group and Fufangbiejiaruanganpian group was lower obviously than that in normal control group（P〈0.01）.There was no difference between Danhong Rangan capsule group and Fufangbiejiaruanganpian group on HSC cell proliferation inhibition rate（P〈0.05）.The expression of type Ⅰcollagen and TGF-β1 in Danhong Rangan capsule group and Fufangbiejiaruanganpian group were notably lower than those in normal control group（P〈0.01）. There was no difference between Danhong Rangan capsule group and Fufangbiejiaruanganpian group on the levels of TGF-β1 and type Ⅰcollagen（P〈0.05）.Conclusion Danhong Rangan capsule can obviously inhibit the proliferation of rat HSCs activated,and inhibit the production of TGF-β1 in HSC-T6.So it can inhibit the expression of type Ⅰcollagen in HSC-T6.This may be one of the mechanisms of anti-liver fibrosis.
