摘要
利用高效液相色谱-二极管阵列检测器(HPLC-DAD)建立简单、快速、精确、同时测定厚朴和凹叶厚朴中绿原酸、芦丁、金丝桃苷、槲皮苷、槲皮素、和厚朴酚、厚朴酚的测定方法。色谱分离是反相C_(18)柱,以乙腈-0.1%甲酸为流动相梯度洗脱,在检测波长294 nm处进行分析。在研究的浓度范围内线性良好,r大于0.9995,回收率为98.28%~104.19%。该方法有效地应用于测定24个源于不同产地、不同采集时间的厚朴和凹叶厚朴样品中7个化学成分的测定。根据这7个特征化合物的浓度,利用聚类分析(HCA)及组成分分析(PCA)对这些厚朴和凹叶厚朴样品进行分类和区分,为合理利用和指导中药资源提供了依据。
A simple, sensitive and accurate high performance liquid chromatography with photodiode-array detection (HPLC-DAD) method has been developed for simultaneous determination of chlorogenic acid, rutiu, quercitrin, quercetin, hyperoside, magnolol and honokiol in Mignolia officina!is Rehd. et Wils. and Mignolia officinalis Rehd. et Wils. var. biloba Rehd et Wils. Chromatographic separations were performed on a reverse-phase Cls column with the gradient elution of acetonitrile-0.1% formic acid and the simultaneous detection at 294 nm. Good linear behaviors over the investigated concentration ranges were observed with the values of r higher than 0.9995 for all the analytes. The recoveries measured at three levels varied from 98.28% to 104.19%. The validated method was successfully applied for the simultaneous determination of the seven chemical constituents in 24 batches of samples collected from different regions or time that were investigated and authenticated as M. officinalis. Hierarchical clustering analysis (HCA) and principal components analysis (PCA) were performed to differentiate and classify the samples based on the contents of the seven characteristic constituents. The blending method proposed was demonstrated to be very useful in saving resources and in guiding rational herb use.
