摘要
目的研究神经节苷脂对脑水肿大鼠水通道蛋白4表达的影响。方法建立大鼠重度颅脑损伤模型,随机分成正常组、模型组和治疗组。考察大鼠伤后不同时间点神经节苷脂对脑含水量和脑组织AQP 4mRNA表达水平的影响。结果模型组和治疗组大鼠脑组织含水量在造模4 h后逐步上升,治疗组在24 h到达峰值后下降,72 h大鼠脑组织含水量值与正常组接近,而模型组大鼠脑组织含水量一直升高,72 h含水量高达80%。两组大鼠造模后1 h后AQP 4 mRNA表达水平开始上调,逐步增加,治疗组在12 h达峰值,然后表达持续回落,72 h与正常组表达水平接近;模型组在24 h达峰值,且表达维持在高位,明显高于治疗组(P<0.05)。结论神经节苷脂能抑制脑水肿的发生,减轻脑水肿形成。
[Objective] To study the ganglioside of brain edema in rats aquaporin 4 expression. [Methods] The rat model of severe brain injury were randomly divided into normal group, model group and ganglioside treatment group. Study on rats at different time points after injury, ganglioside on brain water content and brain levels of AQP 4 mRNA expression were evaluated. [Results] The model group and the treatment group in modeling the brain water content gradually increased after 4 h. The treatment group decreased after the peak reached in 24 h and water content in brain was close to the normal group 72 h, while the model rats with brain tissue water has increased, and 72 h values of brain water content reached 80% moisture content. Rats in both groups after 1 h, the expression of AQP 4 mRNA level began to rise. The level of treatment group reached the peak 12 h and the expression levels was close to normal group 72 h; the expression level of model group reached the peak at 24 h, and then to maintain at a high level, which was significantly higher than that of treatment group (P 〈0.05). [Conclusion] Gangliosides can inhibit the expression of brain edema and cerebral edema formation.
