摘要
目的:了解芪竹方对细胞的毒作用,了解芪竹方对人脐静脉内皮细胞(HUVEC)刺激增殖后的细胞内蛋白酪氨酸激酶(PTK)活性和P-STAT3表达的影响。方法:建立对比模型组,用CCK-8法检测芪竹方对HUVEC增殖的影响,用ELISA法定量测定细胞中PTK含量,用Western Blot法测定细胞P-STAT3表达。结果:芪竹方对HUVEC正常增殖无影响;HUVEC被酸性成纤维细胞生长因子(aFGF)刺激后,空白组PTK和P-STAT3表达异常,芪竹方组对PTK表达有抑制作用,抑制率达65%,同时对P-STAT3表达有抑制,高浓度的芪竹方组作用与阳性药相当。结论:芪竹方对细胞无毒作用,对细胞的PTK和P-STAT3的异常表达有抑制作用。
Objective:To understand the toxic action of Qizhu Decoction to cells and its effect on activation of endocellular protein tyrosine kinase(PTK)and P-STAT3 after it stimulated proliferation of human umbilical vein endothelial cells(HUVEC).Methods:Control group was established to use CCK-8 method to measure the effect of Qizhu Decoction on proliferation of HUVEC,and ELISA method was used to quantify the content of PTK in cells,and Western Blot method was taken to assay the expression of P-STAT3.Results:There was no significant influence of Qizhu Decoction on normal proliferation of HUVEC.When HUVEC was stimulated by acid fibroblast growth factor(aFGF),the expressions of PTK and P-STAT3 were abnormal in the blank control.Qizhu Decoction had an inhibitory action to expression of PTK and the inhibitory rate was about 65%.It also had an inhibitory action to expression of P-STAT3,while high concentration of the decoction equaled the effect of a positive drug.Conclusion:Qizhu Decoction has no toxic action to cells and exerts an inhibitory action to expressions of PTK and P-STAT3.
